Fertility was expressed as the percentage point deviation of bull non-return rate (PD) from the average fertility of all bulls at an artificial insemination center. Number of services for each bull ranged from 1,074 to 52,820 and PD values, from + 7.7 % to – 6.6 %. CEF from 20 bulls was obtained from vasa deferentia cannulae and separated from sperm by centrifugation immediately after collection. Samples were evaluated by 2-D SDS-PAGE, gels stained with Coomassie blue and polypeptide maps were analyzed by PDQuest software. Protein quantities, defined as the total integrated optical density of the spots, were compared between groups of high (n = 12; PD ≥ 0) and low fertility sires (n = 8; PD < 0) and were also used as independent variables in regression analysis. Proteins were identified by capillary liquid chromatography nanoelectrospray ionization tandem mass spectrometry (CapLC-MS/MS). An average of 118 spots was detected in 2-D maps of the cauda epididymal fluid but we were unable to distinguish any protein that was only expressed in high fertility and or in low fertility bulls. However, the amount of α-L-fucosidase 2 and cathepsin D was 2.3 and 2.4-fold greater (p < 0.05) in high fertility than in low fertility bulls, respectively. Conversely, the intensities of three isoforms (24 – 27 kDa; pI 6.3 – 5.8) of prostaglandin D-synthase (PGDS) were from 3.2 to 2.2fold greater in low fertility sires (p < 0.05). An empirical regression model established that a significant proportion (R = 0.72; p < 0.0001) of the variation in fertility scores (PD values) was explained by the intensities of cathepsin D and one isoform of PGDS (24 kDa; pI 6.3). Thus, multiple proteins present in the cauda epididymal fluid are potential biomarkers of fertility in high use, mature Holstein bulls.

Bromochloroacetic acid exerts qualitative effects on rat sperm: implications for a novel biomarker.
Disubstituted haloacid by-products of drinking water disinfection such as dibromoacetic acid and dichloroacetic acid have been shown to perturb spermatogenesis and fertility in adult male rats. In the present study we sought to establish whether equimolar exposure to bromochloroacetic acid (BCA), a prevalent by-product in finished drinking water, is also capable of disrupting these endpoints, a...

In the present study, the fertilizing potential of semen recovered from slaughtered bulls epididymis was evaluated after cryopreservation, by conventional techniques and flow cytometry methods. The cauda epididymal was dissected and sperm were recovered and evaluated for volume, sperm concentration, and membrane and acrosome integrity using a flow cytometer. Sperm fertility potential was tested...
متن کاملMOLECULAR REPRODUCTION AND DEVELOPMENT 74:214–222 (2007) Proteins of the Accessory Sex Glands Associated With the Oocyte-Penetrating Capacity of Cauda Epididymal Sperm From Holstein Bulls of Documented Fertility
We previously reported that accessory sex gland fluid (AGF) from high fertility (HF) bulls influenced the oocyte-penetrating capacity of cauda epididymal sperm from low fertility (LF) bulls, based on in vitro fertilization (IVF) assays. The present study determined if AGF proteins were associated with these effects. Nineteen IVF assays with 12 bulls were grouped as follows. Group I (n1⁄48): ass...

The cryopreservation of epididymal sperm is important to preserve genetic material from valuable deceased males. This study evaluated the viability of sperm samples from eight stallions under three conditions: (1) collected using an artificial vagina (EJ-0h), (2) recovered from the epididymal cauda immediately after orchiectomy (EP-0h), and (3) recovered from the epididymal cauda after 24h of s...
متن کاملDifferential proteomics leads to identification of domain-specific epididymal sperm proteins.
The alteration in the protein signatures of the testicular sperm during its epididymal sojourn makes it functionally competent for successful fertilization. The present study was undertaken to identify the proteins acquired on its 2 domains, that is, the head and the flagellum, during the epididymal transit using a differential proteomics approach. Testicular sperm proteome was compared with ca...
